[目的] 研究胶体金免疫层析法测定生乳中抗生素——氯霉素、氟喹诺酮类含量读数结果的不确定度评定。[方法] 氯霉素含量测定采用“易瑞乳中氯霉素单检快速检测试纸条”、氟喹诺酮类含量测定采用“易瑞乳中氟喹诺酮类抗生素快速检测试纸条”,通过分析不确定度来源,构建数学模型,分析、评定模型每一部分,合成相对标准不确定度,并进一步扩展,得出生乳中抗生素——氯霉素、氟喹诺酮类含量的扩展不确定度。[结果] 均取置信度为95%时,得到生乳中抗生素含量读数结果分别为:X(氯霉素)=3.20±0.059 6,取k=2,扩展不确定度U=0.059 6、X(氟喹诺酮类)=3.00±0.059 4,取k=2,扩展不确定度U=0.059 4。[结论] 试验过程的不确定度主要来源于样品重复性、取样量、试纸读数仪、结果计算引入不确定度。该方法可为免疫分析法(胶体金免疫层析法)测定生乳中抗生素含量的不确定度评定提供参考。
[Objective] To evaluate the uncertainty of the readout results of antibiotic content (chloramphenicol and fluoroquinolones) in raw milk using Colloidal Gold Immunochromatographic Assay. [Method] The "Yirui Chloramphenicol Single Test Rapid Detection Test Strip" [1] was used for chloramphenicol content measurement, and the "Yirui Fluoroquinolones Antibiotics Rapid Detection Test Strip in Milk" [2] was used for fluoroquinolones content measurement.By analyzing the sources of uncertainty, a mathematical model was constructed. Each part of the model was analyzed and evaluated, and the relative standard uncertainty was synthesized and further expanded to obtain the expanded uncertainty of antibiotic content (chloramphenicol and fluoroquinolones) in raw milk. [Result] At a confidence level of 95%, the readout results of antibiotic content in raw milk were: X(chloramphenicol) = 3.20±0.059 6, with k=2 and an expanded uncertainty U=0.059 6; X(fluoroquinolones) = 3.00±0.059 4, with k=2 and an expanded uncertainty U=0.059 4. [Conclusion] The uncertainty in the experimental process mainly originates from sample repeatability, sampling volume, test strip reader, and uncertainty introduced in result calculation. This method can provide a reference for uncertainty evaluation of antibiotic content in raw milk measured by immunoassay(colloidal gold immunochromatography).
