Identification and Drug Resistance Analysis of Staphylococcus aureus from Dairy Goat Mastitis--A Case Study of Liaocheng, Shandong Province

Abstract: [Objective] In order to understand the distribution of Staphylococcus aureus(S.aureus) and its drug resistance genes from dairy goat mastitis. [Method] 178 milk samples were collected from some dairy goat farms in Liaocheng city,Shandong Province,and S.aureus was isolated and identified by bacterial culture,plasma coagulase test and PCR biological technology,and the drug resistance of the isolated strains was further analyzed. [Result] 23 strains of S.aureus were identified,and the detection rate was 12.92%.The results of drug sensitivity test showed that the isolated strains were resistant to penicillin,doxycycline,streptomycin,clarithromycin and tetracycline.PCR detection of drug resistance genes showed that 10 kinds of drug resistance genes were detected,including β-lactam drug resistance gene mecA,macrolide drug resistance genes ermA,ermB,ermC,tetracycline drug resistance gene tet,aminoglycoside drug resistance genes AAC(6')-1A,APH(2'')-1B,ant(4',4'').β-lactam resistance gene mecA, macrolide resistance genes ermA, ermB, ermC, tetracycline resistance gene tet, aminoglycoside resistance genes aac(6')-Ⅰa, aph(2'')-Ⅰb, ant(4',4''), aph(3')-III, and sulfonamide resistance gene sul1 had average carriage rates of 30.43%, 31.88%, 43.48%, 21.73%, and 8.70%, respectively.All strains carried at least 2 kinds of drug resistance genes and up to 7 kinds of drug resistance genes,with the largest number of strains carrying 4~5 kinds of drug resistance genes. [Conclusion] The results suggested that there was a risk of S.aureus epidemic transmission in Liaocheng city dairy goat farms,Shandong Province,and the strains showed different degrees of drug resistance,and the drug resistance gene carrying rate was high,which should be paid attention to and strengthened monitoring and prevention.
