微生物试管法测定叶酸含量的方法研究

摘要

叶酸是鼠李糖乳杆菌生长的必需营养素,微生物法检测叶酸是利用鼠李糖乳杆菌生长速度与叶酸的含量相关性建立的。鉴于微生物法试验周期长,菌种传代保藏比较繁琐,该法是基于《GB 5009.211—2014食品安全国家标准 食品中叶酸的测定》方法,改进了乳酸杆菌接种液的制备方法和叶酸培养液的培养时间。本文通过分析方法的标准曲线的线性、精密度、准确度和方法稳定性来验证优化方法的适用性。该方法标准曲线线性关系良好(R2>0.99),回收率>90%,精密度和准确度好,操作简便,缩短了试验周期,提高了检测效率,为改进叶酸检测方法提供了依据。

Abstract

Folic acid is an essential nutrient for the growth of Lactobacillus rhamnosus,The microbial assay of folic acid was established by using the correlation between the growth rate of Lactobacillus rhamnosus and the content of folic acid.In view of the long test cycle of microbial method and the cumbersome preservation of bacteria,this method is based on the method of GB 5009.211-2014 National Food Safety Standard Determination of Folic acid in Food,improving the preparation method of lactobacillus inoculant and the culture time of folic acid culture liquid.This paper verified the applicability of the optimization method by analyzing the linearity of the standard curve,precision,accuracy and stability of the method.This method has good curve relation (R2 > 0.99),good recovery > 90%,good precision and accuracy,simple operation,shortens test period,improves detection efficiency,and provides a basis for improving the detection method of folic acid.
