Determination of Vitamin D2 and D3 in Milk Powder by Semi-preparative HPLC

Abstract: Purpose: In order to accurately determine the content of vitamin D in milk powder and simplify the pretreatment process of national standard method, a method for the determination of vitamin D2 and D3 in milk powder was established. After saponification,n-hexane extraction and concentration,the sample and the standard solution were purified by semi-preparative NP-HPLC,analyzed by RP-HPLC,quantified by external standard method. Result: There was a good linear relationship between vitamin D2 and D3 in the range of 0.01~0.50 μg/mL. The detection limit was 0.7 μg/100 g and the quantitative limit was 2.0 μg/100 g. The results of six determinations of vitamin D2 and D3 were within the range of characteristic values,and the RSD of reproducibility was 1.07% and 2.38%. The recoveries of vitamin D2 and vitamin D3 were 89.50%~98.70% and 88.17%~95.90%. The relative standard deviations were not higher than 8.81%. Conclusion: The determination of vitamin D in a variety of commercial milk powder showed that the simple and accurate method can be provided a basis for the determination of vitamin D2 and D3in milk powder.
