Determination of 1,3-dioleoyl-2-palmitoylglycerol in Infant Formula Milk Powder

Abstract: [Objective] Establish a gas chromatography method to determinate the content of 1, 3-dioleoyl-2-palmitoylglycerol in infant formula.[Method] The infant formula samples were dissolved in warm water, saponified with ammonia, extracted oil with ethyl ether and petroleum ether, dissolved in n-hexane, separated by DB-1HT capillary column, determined the content of 1, 3-dioleoyl-2-palmitoylglycerol by comparison with standard triglyceride solution, and quantified by internal standard method.[Result] The quantitative limit of method was 0.008 mg/100 g with good linear range.The recoveries of low, medium and high standard addition levels were 81.1% ~ 98.4%, and the relative standard deviations were 2.89%~ 7.88%.RSD of 1, 3-dioleoyl-2-palmitoylglycerol in the matrix samples of infant formula sheep milk powder and cow milk powder was determined to be less than 3.03%.[Conclusion] This method has the advantages of simple operation, short analysis period, high sensitivity and good repeatability. It is suitable for the determination of 1, 3-dioleoyl-2-palmitoylglycerol content in infant formula cow milk powder and sheep milk powder, and has ideal quantitative effect and application prospect.
