牛羊源硬蜱种类鉴定及携带梨形虫的检测与分析

Species Identification of Hard Ticks from Cattle and Sheep and Detection and Analysis of Carrying Piroplasma

摘要: [目的]本研究旨在了解山东省沂南县牛羊体表硬蜱的种类及硬蜱携带无形体病原体属情况。[方法]选取沂南县部分乡镇为采样地点,从牛羊体表采集硬蜱,通过形态学特征对其进行初步鉴定,随后以线粒体cox1基因为靶标对其进行分子鉴定,并以18S rRNA基因为靶基因,利用PCR法对蜱虫携带梨形虫病原体的情况进行调查。[结果]共采集获得硬蜱338 只,其中,长角血蜱219 只,微小扇头蜱119 只;5 个长角血蜱cox1基因序列与已知长角血蜱同源性最高(99.7%~99.9%);5个微小扇头蜱与已知微小扇头蜱相似性最高(99.3%~99.6%),同属分支B。PCR鉴定发现硬蜱巴贝斯属和泰勒属平均检出率分别为7.10%(24/338)和3.25%(11/338)。[结论]长角血蜱和微小扇头蜱为沂南县牛羊体表优势硬蜱,且硬蜱携带梨形虫情况较常见,当地相关部门和养殖企业(户)应加强牛羊硬蜱的检测与防控。

Abstract: [Objective] This study aimed to explore the tick species on the body surface of cattle and sheep in Yinan county Shandong Province,and detect the prevalence of parasitic tick-borne pyriformis..[Method] Hard ticks were collected from the surface of cattle and sheep,and their morphological characteristics were preliminarily identified.Subsequently,molecular identification was carried out using the mitochondrial cox1 gene,and the 18S rRNA gene was used as the target gene to investigate the prevalence of parasitic tick-borne pyriformis.[Result] A total of 338 ticks were collected,including 219 longhorn blood ticks and 119 microfan-headed ticks. The COX1 gene sequences of five longhorn blood ticks showed the highest homology (99.7%~99.9%) with known species, while five microfan-headed ticks exhibited the highest similarity (99.3%~99.6%) to known species, all belonging to branch B. PCR analysis revealed an average detection rate of 7.10% (24/338) for Babesia and 3.25% (11/338) for Tyleria in longhorn blood ticks. [Conclusion] The results indicated that longhorn blood ticks and microfan-headed ticks are the dominant surface ticks on cattle and sheep in Yinan County,with common carriage of Paragonimiasis. Local authorities and livestock farming enterprises should enhance tick detection and control measures.
