奶酪棒中黄曲霉毒素M1含量检测方法的研究与优化

Research and Optimization of the Detection Method for Aflatoxin M1 Content in Cheese Sticks

摘要: [目的]针对奶酪棒成分复杂、传统检测方法操作繁琐且易引入有害化学品的问题,本研究旨在优化奶酪棒中黄曲霉毒素M1(AFM1)的检测方法,核心目标为减少有害化学品使用、简化操作步骤并提升检测准确性与效率。[方法]以甲醇作为提取介质替代传统二氯甲烷,通过优化前处理流程(样品称量、提取、浓缩复溶),结合Prognosis酶联免疫试剂盒进行检测;通过验证定量限、2倍定量限、5倍定量限等不同浓度加标样品,系统评估方法的回收率、相对误差及操作效率。[结果]方法优化后,AFM1回收率稳定在70%~120%,符合《GB/T 33411—2016 酶联免疫分析试剂盒通则》要求;相对误差控制在±25%以内,满足《SN/T 2775—2023 商品化食品检测试剂盒评价方法》标准;前处理时间从传统方法的70 min缩短至25 min,操作步骤从4 步简化为2 步。[结论]优化后的甲醇提取-酶联免疫检测方法,兼具良好的提取效果、检测准确性与高效性,能满足乳品企业批量检测需求,为奶酪棒中AFM1含量检测提供精准可靠的技术支撑。

Abstract: [Objective] Aiming at the problems of complex composition of cheese sticks,cumbersome operation of traditional detection methods,and easy introduction of harmful chemicals,this study aimed to optimize the detection method of aflatoxin M1(AFM1) in cheese sticks. The core goals were to reduce the use of harmful chemicals,simplify operation steps,and improve detection accuracy and efficiency. [Method] Methanol was used as the extraction medium instead of traditional dichloromethane. By optimizing the pretreatment process (sample weighing,extraction,concentration and reconstitution),combined with the Prognosis enzyme-linked immunosorbent assay (ELISA) kit for detection; the recovery rate,relative error and operation efficiency of the method were systematically evaluated by verifying spiked samples with different concentrations such as limit of quantification (LOQ),2-fold LOQ and 5-fold LOQ. [Result] In the optimized method,the recovery rate of AFM1 was stably between 70% and 120%,which met the requirements of GB/T 33411—2016 General Rules for Enzyme-Linked Immunosorbent Assay Kits; the relative error was controlled within ±25%,which complied with the standard of SN/T 2775—2023 Evaluation Method for Commercial Food Detection Kits.The pretreatment time was shortened from 70 min of the traditional method to 25 min,and the operation steps were simplified from 4 steps to 2 steps. [Conclusion] The optimized methanol extraction-ELISA detection method has good extraction effect,detection accuracy and efficiency,can meet the batch detection needs of dairy enterprises,and provides accurate and reliable technical support for the detection of AFM1 content in cheese sticks.
