17 味中药及其复方对奶牛乳房炎金黄色葡萄球菌的体外抑菌试验研究

摘要: [目的]探索17 味中药及其复方对奶牛乳房炎金黄色葡萄球菌的体外抑菌效果。[方法]本试验以山东省某奶牛场患乳房炎奶牛乳样中分离和保存的金黄色葡萄球菌作为研究对象,采用药敏试验和试管二倍稀释法,分别确定金黄葡萄球菌对17 味中药的药物敏感性,测定最低抑菌浓度(MIC)和最小杀菌浓度(MBC),以此选取对金黄色葡萄球菌高度敏感的中药两两组合制成双联中药复方,并采用微量肉汤稀释棋盘法测定双联中药复方对金黄色葡萄球菌的MIC,计算双联中药复方的分级抑菌浓度(FIC)。[结果]黄芩、连翘和五倍子3 味中药对金黄色葡萄球菌的抑菌表现为高度敏感;黄芩对金黄色葡萄球菌MIC和MBC的检测结果分别为7.813 mg/mL和15.625 mg/mL;连翘和五倍子2 味中药对金黄色葡萄球菌MIC和MBC的检测结果分别为15.625 mg/mL和31.25 mg/mL;黄芩+连翘、黄芩+五倍子、连翘+五倍子3 种双联中药复方对金黄色葡萄球菌MIC的检测结果均为15.625 mg/mL,且双联中药间均表现无关作用。[结论]对奶牛乳房炎金黄色葡萄球抑菌效果较好的单味中药组成复方后,其抑菌效果未必增强。

Abstract: [Objective] To explore the antibacterial effect of 17 Chinese herbs and their compounds on Staphylococcus aureus in dairy cow mastitis in vitro. [Methods] Staphylococcus aureus isolated and preserved from milk samples of cows with mastitis in a dairy farm in Shandong Province was used as the study object. Drug sensitivity test and tube double dilution method were used to determine the drug sensitivity of Staphylococcus aureus to 17 traditional Chinese medicines, and the minimum inhibitory concentration (MIC) and minimum bactericide concentration (MBC) were determined. In this study, a combination of two Chinese herbs highly sensitive to Staphylococcus aureus was selected to make a compound compound. The MIC of the compound against Staphylococcus aureus was determined by microbroth dilution checkerboard method, and the graded inhibitory concentration (FIC) of the compound was calculated. [Results] Scutellaria baicalensis, forsythia and gallnut were highly sensitive to Staphylococcus aureus. The results of MIC and MBC detection for Staphylococcus aureus were 7.813 mg/mL and 15.625 mg/mL, respectively. The results of MIC and MBC detected by forsythia and gallnut were 15.625 mg/mL and 31.25 mg/mL, respectively. The results of the detection of MIC for Staphylococcus aureus were 15.625 mg/mL for scutellaria baicalensis + forsythia, scutellaria baicalensis + gallnut and forsythia + gallnut, and there was no correlation between the two compounds. [Conclusion] The antibacterial effect of staphylococcus aureus on dairy cow mastitis may not be enhanced after the composition of single Chinese herbs.
